Acetaminophen overdose causes kidney injury via oxidative stress and apoptosis induction. Coenzyme Q10 (CoQ10) has several effects such as antioxidant, anti-inflammation and anti-apoptotic properties. The aim of this study was to determine the protective effect of CoQ10 on acetaminophen-induced kidney injury in rats. In this study, 40 male Wistar rats were randomly divided to five groups (n=8) including control, CoQ10 10 mg/ kg, acetaminophen, acetaminophen+CoQ10 5 mg/kg and acetaminophen+CoQ10 10 mg/kg. Animals initially received a single dose of acetaminophen (700 mg/kg), then CoQ10 treatment was given as two i.p. injections at 1 and 12 h following acetaminophen administration. Then, animal were sacrificed and Malondialdehyde (MDA) concentration, Glutathione Peroxidase (GPx) and Superoxide Dismutase (SOD) activity levels were measured in the kidney tissue. The western blotting analysis was used to determine the Bax, Bcl-2 and cleaved caspase-3 proteins expression level in the kidney tissue. CoQ10 (10 mg/kg) significantly increased the GPx and SOD activities and also reduced the MDA concentration in the acetaminophen+CoQ10 10 mg/kg group versus acetaminophen group (P<0.05). Also, CoQ10 significantly decreased the cleaved caspase-3, Bax proteins expression level in the acetaminophen+CoQ10 10 mg/kg group versus acetaminophen group (P<0.05). This study showed that CoQ10 can ameliorate the acetaminophen induced acute renal injury at least via suppressing oxidative stress and apoptosis parameters.
