Enhancement of anti-cancerous effect of Resveratrol by conjugating with ZnO nanoparticles

Abstract:

In this study, ZnO Nanoparticles (NPs) conjugated with a well-known anti-proliferative and chemo preventive trans- Resveratrol (RSV) has been designed, characterized and found to be a potential drug for ovarian cancer treatment. Nanoconjugate (RSV-ZnO) has been characterized by FTIR, Raman scattering and Transmission Electron Microscopy (TEM). Picosecond-resolved fluorescence studies of RSV-ZnO nano-conjugate reveal efficient electron migration from ZnO NPs to RSV, eventually enhancing the ROS activity compared to free RSV. Various studies including MTT assay, JC-1 staining method and apoptosis studies on ovarian cancer (PA1) cell lines reveals the RSV-ZnO nano-conjugate to be more effective in cancer cell death in comparison to free RSV. For toxicity evaluation of RSV-ZnO nanohybrid, female Wistar rats (8-10weeks) were randomly divided into 4 groups (n=6) where in group I as vehicle treated control. Group II and Group III and Group IV received 50,100,200 mg of Free RSV and RSV-ZnO for a period of 28 days. RSV-ZnO nano-conjugate did not result in any mortality of animals neither did they exhibit any clinical sign of toxicity. Last part of my work contain efficacy of the drug on DMBA induced rat, we choose a different method to induce epithelial tumors by Intraperitonial Injection (IP) of DMBA. Rats were treated for three days (Day1, Day7 and Day14) during a two week period by intraperitoneal injection of DMBA in corn oil at doses of 50 micrograms/kg day (150 micrograms/kg total dose). After treatment with our drug (dosage of 100 mg/kg bw per week during three weeks study) we have found very positive result with lower expression level of cancer marker (CA125, CK7, Vimentin).
