Abstract:

Survival strategies of extremophiles are unique and as it is least explored; biological potential of extremophilic bacteria is something exclusive to characterize. Geothermal springs are among one of the least explored restricted ecosystem on earth. Water sample was collected from a basic hot spring of West Bengal, India and is not famous for the tourists also. The hot spring is relatively virgin hot spring target and surface temperature is 84 ÂșC. One of the isolated thermophilic bacterium is Bacillus anthracis. Among the halophilic extremophiles; Southern Ocean is a target for researchers as deep southern ocean microorganismâ??s habitat and potential is uniquely different. A Bacillus stratosphericus was isolated from deep Southern Ocean (Indian Sector) that is extreme UV-B tolerant. Isolated samples were characterized using fluorescence, scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Production of exopolysaccharide (EPS) by the isolates was optimized. For morphological characterization of the EPS, Congo red staining, SEM and atomic force microscopic (AFM) studies are done. Elemental analysis, X-ray diffraction (XRD), Fourier Transformed Infrared spectroscopy (FTIR), 1H Nuclear Magnetic Resonance (NMR) Spectroscopy, Thermogravimetric analysis, Differential Scanning Calorimetry, rheological studies were performed for physical characterization of EPSs. Maximum EPS production for Bacillus anthracis was 7.66 g/l with ethyl acetate solvent; whereas maximum EPS production for Bacillus stratosphericus was 11 g/l with ethanolic solvent. As the UV-B exposure time was increasing to Bacillus stratosphericus, production of EPS was also surprisingly increased. As the AFM study revealed nanoparticle like homogenous, sharp and equally distributed molecular structure of the EPSs, EPSsilver nanoparticle conjugates were prepared and it showed promising antimicrobial activity against pathogenic bacteria like- Staphylococcus aureus, Salmonella Typhi and Escherichia coli and different pathogenic fungal strains. EPS-silver nanoparticle conjugateâ??s effective dosage was optimized and is characterized by SEM, XRD and TEM study. Impregnation of AgNO3 on EPS surface is observed. Both the extremophiles showed its beneficial activity on mankind and future studies are needed for production betterment.
